Reproduction and growth of the deep-sea brittlestar ophiura-ljungmani (lyman) ArchiMer

Reproductive biology and population dynamics of Ophiura Ijungmani were studied by measuring disk diameters and by examining oocyte size-frequencies and fecundity from a sequential seasonal series of large samples from a 2900 m deep Permanent Station in the Rockall Trough (Northeast Atlantic). Egg size (max. 90 !lm) and fecundity (up to 57000 eggs per individual) suggests indirect planktotrophic development, while a clear annual cycle in gametogenesis, with probably synchronous spawning in JanuaryFebruary, confirms a marked seasonality that is also seen as an annual, summertime set of postlarvae on to the bottom. Electrophysiological measurements and related cytological controls were done on immature and submature oocytes of Homarus gammarus . By chromatography (HPLC) and radio-immunoassays (RIA) it was established that submature oocytes displayed different ecdysteroids. Furthermore, the structure of the stalk (funiculus) for the laid eggs attachment was analyzed. it consisted of the two superimposed vitelline envelopes which were likely modified by the cortical reaction exudate during the fertilization process. In addition, the successive secretory phases, regarding to the embryonic envelopes were evidenced. Finally, the ecdysteroid titers of embryonic stages were measured.

The lipid content of spawned oocytes of Pecten maximus was studied by image analysis of semi -thin histological sections and a lipid index was defined. The female gametes were obtained by thermic stimulations of mature adults scallops from different populations after out of season conditioning or sampled in situ during the spawning season. After fertilization, the larval development was followed. The results reveal a large variability in lipid content of normal oocytes. The lipid content can be regarded as homogeneous inside a spawning but the heterogeneity is greater for wild scallops than for conditioned ones. PRODUCTION OF OVOGONIA BY GERMINATIVE EPITHELIUM OF THE OVARY AT SOME MAMMALIA AND BIRDS Ciência Rural

The studies were effected on ovaries from sows, cows, bitches, dous and hens. Our investigations revealed the generation of cellular rows of germinative epithelium into the cortex not only during the embrionary period but also in the adult ovary. The histological investigations revealed an intense proliferative process of the germinative epithelium of adult ovary which generated groups of ovogonia under the lamina basalis. From ovogonia developed oocytes and then ovarian follicles. The cause for the proliferation of ovary germinative epithelium could be: the multiple folliculary dehiscences, the premature degeneration of ovarian follicles and formation of the corpus luteum, phenomena that reduce continually the number of embrionary ovogonia.

An in vitro zona-free hamster oocyte penetration test was utilized in 24 trials in order to evaluate the capacitation media used for ram sperm. A pool of fresh semen was collected from three crossed breed rams. Two semen drops were washed by centrifugation and incubated in high ionic strength treatment (HIS) or in a defined medium with HEPES, on heat ewe serum and heparin. After the incubation to promote capacitation, simplified triple-stain technique was used to evaluate the spontaneous acrosome reaction of the capacitated sperm. Ultrastructure of ovarian follicular epithelium of the Amazonian fish Pseudotylosurus microps (Teleostei: Belonidae): morphological and histochemical characterization of the intercellular deposits BJB

The present paper reports the presence of great quantities of electrondense intercellular material in the follicular epithelium of P. microps. The material apparently is uptaken from circulation and enter the follicle through the intercellular spaces accumulating in the epithelial median-apical intercellular spaces and in perioocytic space. The accumulation starts in the early growth of the primary oocyte and proceed until vitellogenesis. The possible chemical nature and function of the deposits are discussed.

The present study was conducted to clarify the difference in the color of the cytoplasm in immature follicular oocytes from prepubertal and adult minke whales. The four lipid contents (triglyceride,total cholesterol phospholipids and non-esterified fatty acids) in vitrified immature oocytes from prepubertal and adult minke whales,and also in fresh and vitrified immature porcine oocytes,were measured. The lipid contents in vitrified-warmed minke whale oocytes were similarly high compared with those in vitrified-warmedp orcine oocytes. Objective: To disclose a new biotechnique, listing some of its aspects and addressing its main stages, isolation, cryopreservation and culture of preantral follicles. Data Sources: The information was obtained from CAB, MEDLINE, PUBMED, Science Direct and CAPES Portal. Data Synthesis: The preantral follicles store about 90% of the oocytes within the ovary. The biotechnique described is based on recovering immature oocytes retained inside preantral follicles before atresia takes place. This paper describes morphological characteristics of the ovarian germinative cells of the Scinax fuscovarius (Lutz, 1925). The ovary is organized in primordial germinative cells (oogonia) and follicular structures (ovarian follicle) - oocytes surrounded by follicular cells. Oogonia: their nests are peripherically localized, containing cells with large and oval nucleus. The reproductive biology of Ramnogaster arcuata inhabiting waters of the Rio de la Plata estuary (Argentina - Uruguay) was studied by using macroscopic and histological analysis of ovaries. R. Arcuata specimens were collected from two research cruises of the National Institute of Fisheries Research and Development (INIDEP) carried out in June 1998 and June 1999. Use of a permanent infusion pump as vacuum generator for ovum pick-up MV&Z

To disclose an alternative equipment for vacuum generation, which allows for oocyte recovery in quality and quantity compatible with specific vacuum pumps. A continuous infusion pump was employed and aspiration was possible due to the use of the infusion fluid line in the reverse way. The end destined for the infusion bottle was connected to the follicle aspiration tube, while the end destined for the patient’s catheter was kept free. The results were similar to those obtained with specific follicle aspiration equipment’s. The continuous infusion pump can represent a viable alternative for recovery of bovine oocytes in vivo.

Vacuum-cooled liquid nitrogen increases the developmental ability of vitrified-warmed bovine oocytes Ciência Rural

The objective of this study was to determine the effects of vacuum-cooled liquid nitrogen on the development of vitrified immature (germinal vesicle stage; GV) and mature (metaphase II; MII) bovine oocytes after re-warming. Liquid nitrogen was exposed to either atmospheric pressure or to a vacuum (300mm Hg for 45sec); the latter decreased the temperature of the liquid nitrogen to -200°C. Partially denuded oocytes were vitrified either just after selection (GV) or after 22 hours of in vitro maturation (MII) in TCM 199 medium + 10% of estrous mare serum. The ovarian histology, the structural and the ultrastructural characteristics of the folliculogenesis in Didelphis albiventris were described in detail. Recent studies suggest that methatherian mammals have unusual reproductive cycle but there are few informations regarding the marsupials reproductive life. Despite of the opossum folliculogenesis pattern resembles methatherian and eutherian pattern in many aspects, the analysis shows some peculiar features of the oocyte structure and ultrastructure that make available new data on the reproductive biology of marsupials.

Influence of climatic conditions on in vitro production of bovine embryos Arq. Bras. Med. Vet. Zootec.

Temperature and rainfall were analyzed daily during six years to evaluate their influence on in vitro production of bovine embryos. Weekly replications (n=480) were performed on 14,778 ovaries collected at slaughterhouses. Cumulus oocyte complexes (n=19,180) were fertilized with a pool of Bos taurus taurus semen in one incubator with 5% CO2. Presumable zygotes were cultured in gasified plastic bags with 5% CO2, 5% O2, and 90% N2. In the first year, cleavage and embryo yield were 60.3% and 15.6%, respectively, being lower (P<0.05) than in the following years. Since the 1970s, There have been studies of the venom of Latrodectus sp. spiders, in particular the latrotoxin (LTX) of Latrodectus mactans. Many of the studies were aimed at understanding the action of the venom on the muscular system. Now accepted that LTX is able to generate a calcium-permeable membrane pore and modulate the release of synaptic vesicles that activate a receptor and induce cellular changes. Reproductive aspects of the Brazilian snapper Lutjanus alexandrei, were characterized, including a description of the development of oocytes and spermatogenic cells, size at first sexual maturity, and fecundity. A total of 540 fish were analyzed with 250 having their gonads sectioned to allow microscopic evaluation. Six maturity stages were identified for females and males: immature, maturing, mature, spawning, spawned, and resting. Fish standard length (SL) varied from 13.0 to 28.3 cm and sex ratio was 1.6 males: 1.0 females.
